The Study of DNA Origami as Target Nano Delivery Vehicle for Crossing Blood-brain Barrier Based Optical Molecular Imaging 

  

Abstract: Drug treatments of central nervous system diseases often compromised the presence of blood-brain barrier and how to use a safe and effective carrier, neither damages the normal function of the brain, but also increases the permeability in the blood-brain barrier becomes particularly important. We use novel DNA origami biocompatible nanocarriers, which target the brain capillary endothelial cells high expression density lipoprotein receptor. The different shapes nanocarriers are modified with ApoE protein, meanwhile long emission wavelength dye is coupled on the surface. These multifunctional nanocarriers integrate targeting and imaging together. Optical molecular imaging is used to give different DNA origami shapes carriers across the blood-brain barrier efficiency in vivo, real-time assessment. All above lays a solid foundation for targeted follow-central nervous system diseases drug delivery.
